Dr. Martha Connors was a scientist at Empire State University Labs. She is an expert in biology along with her husband Dr. Curt Connors. They have a son Billy Connors who hangs about the lab with them. Her husband Dr. Connors lost his arm in an accident and had a functioning, mechanical prosthetic arm. Her husband was obsessed with uncovering the secrets of reptilian limb regeneration and created a serum made of modified lizard DNA. He injected himself with it frequently to help re-grow his lost arm. In a fight in the Lab between Spider Man and former electrician Max Dillon/Electro, one of the vials was hit by Electro's electricity, amplifying the DNA within the serum. As a result of injecting the altered Lizard serum, Connor's arm re-grew as expected. However, Connors was later fully transformed into the Lizard. The Lizard's could not talk and acted purely on animalistic instincts. The Lizard was extremely aggressive, shown attacking with little or no provocation, and even coming close to biting Billy. An antidote called a gene cleanser, developed by Spider-Man, Eddie, Gwen, and Martha managed to restore back to his normal self. When the alien symbiote was found on Colonel. John Jameson's Shuttle, it was taken to the ESU Labs to be studied by the Connors. The pair was ecstatic about the discovery and given new hope to their lives and career, but after the robbery in which it got attached to Spider-Man, they were distraught over the loss. Later the science department gained Dr. Miles Warren as a new member as Norman Osborn funded the research. Dr. Warren had been conducting mutagenic experiments on human test subjects; her husband threatened to inform the school board only for Dr. Warren to blackmail him with his own lizard experiments. Later after all their troubles and losses, Martha and Curt decided to leave with Billy for the Florida Everglades to escape Warren's blackmail.
