Leonardo was a thirteen-year-old anthropomorphic mutant who lived in a secret, subterranean home hidden within the sewer systems beneath New York City. Alongside his three biological brothers, he was raised and trained from infancy under the strict guardianship of Master Splinter, a giant, intelligent rat who educated them in the deadly ways of ninjutsu. After completing a successful late-night skirmish against a violent street gang called the Purple Dragons, Leonardo and his brothers returned to their lair, where Splinter deemed them finally prepared to learn the truth of their existence. He learned that their mutation was caused by a strange, glowing canister of cosmic ooze that dropped into the sewers twenty years prior, and that their master’s ultimate purpose was to seek blood retribution against a ruthless Japanese assassin known as the Shredder. Accepting his role in this dangerous family vendetta, Leonardo helped deliver a formal combat challenge to the villain's headquarters, leading his brothers into a brutal rooftop confrontation where they successfully overwhelmed the martial arts master and avenged their family line. Personality and attributes
In appearance, Leonardo was a large, heavily built humanoid turtle who possessed an exceptionally athletic, muscular physique and tough, green-skinned reptilian flesh. As a cold-blooded mutant, his physical form was defined by a massive, highly durable protective carapace shell on his back and a segmented plastron plate covering his torso. His hands were adapted into three thick, clawed digits, while his feet featured two large, powerful toes designed for heavy traction and agile acrobatics. Unlike later adaptations that assigned unique, bright multi-colored gear to individual members of the team, Leonardo and his brothers wore entirely identical ninja gear to maintain group uniformity in the shadows. He wore a simple, dark cloth mask wrapped tightly around his head that covered his brow and featured long, flowing ties behind his neck, with cutouts that exposed his narrow, focused white eyes. Among his siblings, he was commonly shown wearing a blue eye mask. His wrists, elbows, and knees were wrapped in thick, protective combat bands, and a wide leather utility strap buckled horizontally across his waist to anchor twin scabbards on his lower back.

Leonardo possessed peak mutant conditioning, Olympic-level athletic capabilities, and masterful execution of silent ninjutsu espionage tactics refined through a lifetime of underground isolation. He exhibited immense, lethal proficiency with traditional Japanese weaponry, specializing specifically in the dual-wielding of razor-sharp katanas to slice through enemy armor, deflect oncoming projectile fire, and execute fatal melee strikes. His heightened senses and physical training granted him near-superhuman spatial awareness and stealth, enabling him to seamlessly blend into the dark city rooftops, scale urban structures, and vanish from sight before human authorities could arrive. Beyond his individual martial prowess, Leonardo demonstrated an innate tactical mind and a deeply disciplined, honorable focus during chaotic encounters. When facing an overwhelmingly superior martial artist, he utilized precise squad coordination, directing his brothers to abandon individual combat in favor of synchronized, long-range group maneuvers to successfully chip away at and neutralize a deadly opponent.

- In Teenage Mutant Ninja Turtles, Leonardo appeared in the setting of the 1987 animated television series where he was voiced by actor Cam Clarke. Leonardo was a pet turtle along with three others who had been bought by a boy, but the boy tripped and the four turtles fell into a sewer drain. At the time, Hamato Yoshi had been hiding in the sewer from his enemy, Oroku Saki, and had become a friend of the rats living down there. Hamato was overjoyed to find four new friends, and welcomed the turtles into his home. At some point after, Hamato noticed the turtles covered in a glowing substance, and he touched the substance himself. The turtles had most recently touched Yoshi, causing them to quickly evolve into humanlike turtles. Hamato, having just been exposed to the rats, became a human rat. Hamato saw the turtles' potential to challenge the one responsible for the mutations to change him back to a human, and trained them in the art of ninjutsu, and naming them after some of his favorite Renaissance artists – Michelangelo, Leonardo, Raphael, and Donatello. Some years after, April O'Neil was giving a news report on a recent break-in at a science lab, and a scientist used witnesses of the thieves and bits of rope to conclude the thieves were most likely ninja. April and her crew went to the next potential science lab robbery, only to moments later be chased by a group of street gang members. April managed to slip into a sewer drain, but the gang members followed. Suddenly, a second group of ninjas appeared and took out the gang. April fainted when she saw Leonardo and the other ninja were turtles. Leonardo and the others took her to their Master Splinter, who explained the Turtles' origin to her. April, however, believed that the Turtles were the thieving ninja, but the Turtles made a deal: they would help her find the one behind the thefts and get her a story, while she would try her best to find the Mutagen creator. She took the Turtles to the surface and bought the Turtles coats and fedoras to keep the Turtles' bodies unrevealed, and went to the scene of the crime, finding only a matchbox with the words "Ninja Pizza" on it. They found and went into a restaurant by the name of Ninja Pizza and ate there.
