History
Slayers were supernaturally empowered women that were chosen to fight against supernatural threats in the world. Their origins were traced thousands of years ago in ancient Africa where a group of tribal elders known as the Shadow Men sought to create an agent to serve their needs. They kidnapped a girl named Seneya where they used their powerful magic to infuse the captive girl with the heart, soul and spirit of a captive demon. This gave her increased strength, speed, agility, reflexes, healing, endurance and psychic abilities allowing her to fight against the forces of evil with her becoming the First Slayer. Thus, she began a line of warriors that were called to fight against the vampires and demons that inhabited the Earth. In time, the descendants of the Shadow Men became the Watchers whose Council were tasked with finding, training and supporting the Slayers. (Episode: End of Days)
In this time, Summers had died but was revived through the use of CPR. (Episode: Prophecy Girl) Her clinical death resulted in a new Slayer being chosen with this being Kendra. This led to the first time in their history where they were two Slayers active at a time. (Episode: What's My Line? Part Two)
After the death of her mother, Buffy Summers came to question her ability to still feel love anymore and that she was becoming a colder person as a result. Giles came to convince her to take part in the Slayer ritual where she could seek out the advice from a guide in a vision quest. (Episode: Intervention)
The mystical rules governing the calling of Slayers changed again in 2003, when Buffy discovered the Scythe, forged by the hidden Guardians, for the Slayer to wield. (Episode: End of Days) In order to defeat the forces of the First Evil, which would have hunted down and murdered all the Potential Slayers and caused an apocalypse without interference from the Slayer, Buffy had her best friend, the witch Willow Rosenberg, use magic to tap into the Scythe's essence and perform the Slayer activation spell, that called multiple Potential Slayers at once, thus ending the legacy of 'one girl in all the world'. Overview
In appearance, the Slayer were female warriors empowered by mystical forces for the purpose of fighting against monstrous forces in the world. (Episode: Welcome to the Hellmouth) According to the First Slayer, the purpose of the Slayer was to bring death to others. (Episode: Intervention)

A Slayer naturally had formidable fighting abilities. Buffy easily fended off multiple vampires at the same time while under a memory loss spell, with no memory of her combat training — relying entirely on her natural Slayer instincts. (Episode: Tabula Rasa)
All Slayers through the ages shares a psychic link, manifested in dreams. (Episode: Damage) A Slayer would frequently dream of herself as a Slayer in another time and place, even before being called, which would hint if they were Potential Slayers. They could also experience prophetic dreams related to upcoming crises. (Episode: Welcome to the Hellmouth)
A special ritual could be conducted by the Slayer to receive guidance in times when they felt lost in their purpose. The location of the site itself was a sacred place that was a guarded secret. In fact, the Slayer was expected to go to the site themselves and could not be accompanied by a Watcher. Instead, the Watcher had to perform a ritual to transfer their guardship of their charge temporarily to a guide. (Episode: Intervention)
Some Slayers were known to gather an 'emergency kit' that was a Slayer heirloom which should have been passed on to the next Slayer. (Episode: Get It Done)
One handbook was Vampyr that was given by a Watcher to their charge to instruct them on how to operate as the Slayer. (Episode: What's My Line? Part Two)
According to the Slayer Handbook, the Slayer was supposed to separate herself from civilization for security. (Episode: What's My Line? Part Two)

Comics
- In Fray (2002), the Slayers appeared in the setting of the Dark Horse comic that was set in the Buffy-Angel universe. Set in the 23rd century, a new Slayer was shown to operate in this era with this being a woman named Melaka Fray.
- In Buffy the Vampire Slayer Season Eight (2007), the Slayers appeared in the comic continuation of the live-action television series. One of the newly activated Slayers was a girl named Satsu who was inducted into the Slayer Organization where she entered into a brief sexual relationship with her mentor Buffy Summers. Afterwards, she came to lead her own Slayer team that operated in the Tokyo area.
